The Merck Index is a compilation of 10,000 of the most scientifically recognized drugs, pharmaceuticals, organic chemicals, laboratory reagents, and naturally occurring substances. However, you will find the constituents of the Hawaiian Noni polysaccharide (glucuronic acid, galactose, arabinose, and rhamnose).

Myth #4: The "footprint" of Tahitian Noni juice proves that it contains more beneficial ingredients than any other Noni juice.
The Truth:
Where do these "footprints" (or chromatograms) come from? The footprints were analyzed by reversed phase on a high-pressure liquid chromatograph with ultraviolet detector, or HPLC-UV. The Noni juice is filtered and then injected into an "analytical column" (a tube about 1 cm in diameter filled with a packing material). A pump helps the sample move through the column. Different constituents of the juice separate as they meet the packing material. Lighter, more soluble constituents move more quickly through the column and heavier, less soluble constituents move more slowly, reaching the end of the column at a later time. At the end of the tube, a detector analyzes each constituent as it leaves the column. In this case, UV light hits the sample and the amount of UV light absorbed by each constituent is recorded. The more UV light is absorbed, the higher the "peak" in the resulting footprint.
What do the "footprints" mean? Because they do not identify the components corresponding to each peak on the "footprints," the graphs are meaningless. Why aren't the active components identified on the graphs? It is impossible to obtain the same footprint every time a mixture is analyzed, especially when a complicated sample with many constituents (such as Noni juice) is analyzed. For instance: The amount of time it takes for a constituent to pass through the analytical column can vary from one column to another identical column. Each time a column is used, it takes a little bit longer for the sample to pass through. (In the industry, it is said that the "elution" or "retention" time increases.) To accurately describe each juice, the same tests would have to be performed many times and the data from each analysis synthesized.
Why is using a UV detector potentially misleading? Different substances absorb UV light in different ways and substances that absorb the most UV light (those with the highest peaks on the graphs) are not necessarily the most beneficial constituents. A large peak in the footprint may indicate a small amount of a substance that absorbs UV light well and a small peak may indicate a large amount of a substance that does not absorb UV light well. Substances absorb some UV wavelengths better than others do. Thus, at one wavelength Substance A may give a tall peak while Substance B gives a short peak and at another wavelength Substance B would have a taller peak. Some substances absorb no UV light and are therefore undetectable with the UV detector. Thus, the most beneficial ingredients might not even show up on a graph made with a UV detector.
What are the conditions under which each of these graphs was formed? The same HPLC equipment with a different detector, called a mass spectrometer, identifies the mass and quantity of each constituent as it leaves the column. With this equipment, problems with elution time would be irrelevant. Peak detection would depend only upon the amount of each constituent contained in the sample. Each constituent would be easily identified.
